New research finds 51% of U.S. drug launches exceeded Wall Street revenue expectations in 2025, with oncology products leading the way
WALTHAM, Mass., Oct. 5, 2026 /PRNewswire/ — Trinity, the leader in integrating human and artificial intelligence in life sciences commercialization, today announced the release of its 5th Annual Trinity Next Gen Launch Report, revealing that the U.S. pharmaceutical and biotechnology industry delivered its strongest launch performance since Trinity began tracking new drug launches in 2020, despite significant regulatory, economic and market uncertainty.

The report, titled 2025 Launch Paradox: Ambiguity, Overperformance, and What Standouts from the Class of 2025 Teach Us About Launch Excellence in Uncertain Markets, analyzed 36 U.S. pharmaceutical and biotech launches in 2025. The findings show that 51% of launches exceeded Wall Street consensus revenue expectations, significantly outperforming the historical four-year average of 39%.
The strong performance came amid a challenging environment for the life sciences industry, including drug-pricing policy uncertainty, funding pressures at the FDA and NIH, volatile capital markets, and heightened merger-and-acquisition activity. Despite these headwinds, pharmaceutical companies demonstrated an increased ability to execute successful commercial launches and accelerate patient adoption.

Key Findings from the 2025 Next Gen Launch Report
Among the report’s most significant findings:
- 51% of pharmaceutical launches exceeded revenue expectations. Of the 36 U.S. launches analyzed, 51% generated at least 110% of Wall Street consensus revenue forecasts, compared with the historical four-year average of 39%. An additional 11% performed in line with expectations, while 38% underperformed.
- Market volatility created additional challenges. U.S. biopharma M&A activity increased approximately 2.5 times, reaching $113.3 billion in 2025 compared with $46 billion in 2024, while biotechnology equity markets experienced significant fluctuations.
- Oncology launches demonstrated exceptional performance. An impressive 90% of oncology launches exceeded Wall Street revenue forecasts, with several products generating more than 700% of first-year consensus expectations.
- Commercial execution improved despite fewer first-in-class therapies. The number of first-in-class launches declined 39% year over year, from 34 in 2024 to 21 in 2025, suggesting that improved launch performance was driven in part by stronger commercialization strategies rather than scientific innovation alone.
- First-time launch companies continued to close the performance gap. Among companies launching their first commercial product, 63% met or exceeded revenue expectations in 2025, continuing a two-year trend of improved performance.
What the Most Successful Drug Launches Did Differently
Beyond analyzing industry-wide performance, Trinity’s report examines three standout pharmaceutical launches to identify the commercialization strategies that helped companies exceed expectations.
Brinsupri® (Insmed): The first FDA-approved treatment for non-cystic fibrosis bronchiectasis generated $172.7 million in U.S. net revenue in 2025, reaching approximately 385% of Wall Street’s first-year revenue forecast. The launch benefited from prior relationships with HCPs to enable successful early disease awareness initiatives, patient identification efforts and dedicated field access managers that accelerated treatment initiation.
Tryngolza® (Ionis Pharmaceuticals): The first FDA-approved therapy for familial chylomicronemia syndrome generated $108 million in U.S. net sales in 2025, exceeding consensus forecasts by 328%. Its commercial success was supported by initiatives to improve patient diagnosis, tailored genetic testing programs, targeted and tech-enabled patient identification and comprehensive patient-support services.
Attruby® (BridgeBio Pharma): The therapy for transthyretin amyloid cardiomyopathy generated $365.3 million in revenue in 2025, demonstrating how meaningful clinical differentiation, market expansion and reduced access barriers can help a new entrant from a first launch company compete effectively against large pharma and deliver innovation to patients.
Across these case studies, Trinity identified three common strategies that contributed to successful commercialization: establishing meaningful clinical differentiation while investing in market expansion; building demand through early patient identification and effective field execution; and accelerating access through pricing strategies, treatment guidelines and tailored patient-support programs.

As the industry faces continued pricing pressures and increasingly competitive therapeutic markets, that level of preparation and execution will become even more critical.”
Implications for the Next Generation of Pharmaceutical Launches
Trinity’s findings suggest that the pharmaceutical industry’s approach to commercialization is evolving. While clinical innovation remains essential, companies are increasingly distinguishing themselves through their ability to translate scientific advances into effective customer experience and patient access.
The report also highlights the limitations of commercialization strategies in crowded therapeutic categories. In hereditary angioedema, for example, three products were approved in 2025, but none exceeded revenue expectations, illustrating the challenges facing new entrants when existing therapies already address substantial portions of patient need.
Looking ahead, Trinity expects early market preparation, differentiated clinical positioning, patient identification and streamlined treatment initiation to remain central to successful pharmaceutical launches.
